Is it possible to set the online backups of the information store to backup locally to disk and then set the backup of the MDBDATA folder to backup to tape?

You've been able to do this for over eight years. Use ntbackup.exe to backup the Exchange to disk. Then back up the resulting BKF file to tape.

Sorry I wasn't clear when I wrote this.

Basilcally SBS performs an online backup of the information store and then an file system backup of the mdbdata folder. Therefore creating two backups of the information store.

We are running out of space on our tapes so was wondering if I could set the online backup to stay on the local disk and then the file system backup to go onto the tape so we can keep it off site.

This file level backup of the MDBDATA folder is no good to you because it can't capture the open files. You need both types of backups because although the file level backup does not capture the files you think it does, it does capture important ones.
Bottom line: Get more space or store the backups elsewhere, you can backup to a network location.
