When setting up the DAG does all my mailbox servers drives letter needs to be different? eg... if server1 has drive letter G, H does the second servers need to set it's drives a different letter to make DAG work?

No. The proper (and only) way is the complete opposite. For instance if DB1 is e:\Exchange\db1.edb on server1 it must also be e:\Exchange\db1.edb on server2
