Hello!

I have a big problem here. One of the user here is suspected that he is doing telnet to ms exchange and most of the time he is doing a "joke" messages to the employee's. As a Exchange Administrator I need to block this access, I already search on internet for possible solution but no luck. Please help me to fix this threat.

You can't disable "telnet" in the way that you think. The person might be using telnet as a command but it's against port 25 rather than 23.
Stop him by holding him breach of the corporate use policy. If you haven't got one then make one and have him sign it.
Alternatively configure Exchange so that it does not accept connections from any of the workstations on the LAN. You cannot prevent anonymous access because it would prevent anyone on the Internet sending to you.

Expanding upon what Mark said, port 25 is used when servers communicate with each other.  Users connect via client software.  Outlook uses MAPI.  OWA uses HTTP or HTTPS.  Blocking access via port 25 will eliminate this method and force him to use the client software which is easier to track.

You can either block it at the server side, or by disallowing outbound port 25 connections from workstations via the Desktop Firewall.
