Hi there,

If I need to remove the mailbox limit for one user and to give him unlimited storage space temporary, do I just uncheck the "use default setting" in his AD account and leave the other boxes blank?

Yup.
You'll need to give it around 20 minutes to get out of AD (where you made the change) and into Exchange. The information is read and cached on a schedule.
