Is an in-place upgrade from Exchange 2003 Standard edition to the Enterprise version possible and supported? If so, has anybody done it and what should I watch out for, please?

Fully possible and supported.
Main thing is to make sure the backup (before and after) are viable, i.e. you test to alternate hardware before doing anything.

To upgrade the Standard Edition of Microsoft Exchange Server to the Enterprise Edition of Microsoft Exchange Server.

Run the Setup program from he Exchange Server Enterprise Edition compact disc and click the Reinstall button.
