I will be doing a backup prior to migration and have a question on backing up an exchange2000 server. Should the store be unmounted before backing it up and should there be any other processes or services terminated prior to doing the backup.

Did you never make a back up before !?!?!?!?!?

Using NTBACKUP you don't need to stop any service... the users can make a normal use of the mail will you back up it.

Also back up System State and AD. All Exchange Configuration is stored in the AD since Exchange 2000 !
