Relay Agent?

Our environment is like hub and spoke, where many spokes are routing mails to the hub and out to the destinated spoke.  All spokes and the hub are external routes.

We received this error when sending to another spoke:

550 Mailbox unavailable: This site may not be used as a relay agent.

Our test shows that when routing to the hub, without going to a spoke, it's perfectly deliverable with no return error messages (receiver confirmed receiving the message).

What does this error message mean? And how do I go about fixing it?

The DNS structure is this:

Hub:  *.newshub.com
Spoke: *.spoke.newshub.com or *.spoke2.newshub.com

Any help is appreciated.
