I am in the process of decomission exchange01 (I have moved all maiboxes, folders, RUS and so on), but when I shutdown the server for a while SMTP queue rises on exchange02 and monitoring goes into critical status. The only messages I see in queue are system generated messages from exchange02. I am using Default SMTP virtual server and smarthost as mail relay for inbound and outbound mail routing.

Thank you Phil, although I followed both the links but no help. I was able to fix the outbound routing but when I shutdown the exchange01, the SMTP queue on on exchange02 rising and goes to critical state

Did you remember to add Exchange02 as a bridgehead for your outbound SMTP connector?  If not, it will attempt to send all the outbound messages to one of the servers listed (sounds likely Exchange01 is the only one).

Good to see you again .. I did add exchange02 as local bridgehead in my only outbound connector.

Are the messages stuck in a queue destined for Exchange01 (name of the queue)?

If you go into the Exchange System Manager, expand Exchange02 / Protocols / SMTP and open the properties of the Default SMTP Virtual Server.  Go to the Delivery tab and click Advanced.  Do you have anything in the Smarthost field?

Have you restarted the Exchange services on Exchange02 since adding it as a bridgehead?

The only messages get stuck in queue of exchange02 are from exchange02-isadmin@abccorp.com. No I dont have any smarthost listed in Delivery, Advanced tab of Default SMTP Virtual server and yes I restarted all the exchange services on exchange02.

What is the name of the queue in which the messages are stuck?
How many Routing Group connectors do you have defined?

I only have one routing group connector defined. And the name of the queue where messages get stuck is "SMTP Exchange01"
