When I am creating a new user in active directory, the exchange email address in not created automatically. In current environment we have Exchange 2000 with sp3 on windows 2000 sp3 and 2 windows 2000 domain controllers with sp3.
I did not see any errors in the event logs on exchange server and both the domain controllers.

I will appreciate if you can help me resolve the problem.

Sounds like a RUS problem, are the SMTP proxy addresses added to the users when updating RUS manually?
